The antibody was obtained from polyethylene glycol fusion of non-secreting mouse myeloma(P3X63.Ag8.653) cells with spleen cells from Balb/c mouse immunized with human ventricular myosin.

Antibody production : Clinical grade tissue culture supernatant was concentrated and purified to homogeneity by a series of chromatographic steps. Immunoglobulin was then fragmented to F(ab')2 by proteolytic degradation and reduced to Fab' by cysteine.
  
Kit preparation : Pure 3-48 Fab' fragments were "pre-tinned", excess tin removed and antibody fragments fromulated in the presence of technetium reducing agent into sterile hypovials, and lyophilized.
For radiolabelling, vial content is reconstituted with 99mTc freshly eluted from the generator and after 30 minute incubation at room temperature is ready.

Immunologic specificity : The monoclonal antibody is myosin alpha and beta heavy chain specific. On Western blots and in ELISA it reacts with purified heavy chains of atrial and ventricular myosin isolated from human, canine, bovine, porcine, murine, rabbit, and rat cardiac tissues. It binds to A-band of thick filament of cardiac and skeletal muscle myosin as determined by indirect immunofluorescence on cryocuts, and indirect immunoperoxidase staining of Carnoy fixed tissues. It stains human and canine myocytes in culture as determined by indirect immunofluorescence.
  
Cross-reactivity : 3-48 antibody cross-reacts with heavy chains of human myosin isolated from: slow skeletal muscle, fast skeletal muscle, eye muscle, and streptococcal M5 peptide. It does not react with human platelet, smooth muscle myosin heavy chains, myosin light chains, myosin fragments circulating in blood of patients with myocardial infarction, nor does it react with any other human tissue antigens or blood cells.
  
Storage : Store at 4oC.
  
Stability : IgG When reconstituted in aqueous solution IgG remains stable for six months if stored at 4oC. No considerable loss of activity was detected when stored at -70oC over a period of two years.
F(ab')2 When reconstituted in aqueous solution F(ab')2 remains stable for six months
if stored at 4oC. No considerable loss of activity was detected when stored at -20oC over a period of one year.
CVTM Stable for one year when stored lyophilized at 4oC.
