Carcinoembronic antigen (CEA), an oncodevelopmental tumor marker is a fairly typical, high-molecular weight, cell surface glycoprotein over-expressed in adenocarcinomas of entero-dermally-derived organs of the digestive system and in fetal colon. Highly specific anti-CEA mouse monoclonal antibody was produced as a result of a unique immunization protocol.

ANTIBODY CHARACTERISTICS

Production : The antibody was obtained from the polyethylene glycol fusion of non-secreting mouse myeloma (P3X63.Ag8.653) cells with popliteal lymph node lymphocytes from BALB/c mice immunized with the anti-CEA monoclonal antibody C234 mAb.
   
Purification : Tissue culture derived antibody was precipitated with ammonium sulfate and affinity chromatographed on CNBr-activated Separose 4B-C234 mAb.
  
Immunologic class : IgG1 k
  
Immunologic specificity : L12E2 mAb is specific fot the immunizing idiotope of C234 mAb. It binds C234 mAb at the CEA binding site since :
(a) it inhibits the binding of CEA to C234 mAb as well as,
(b) C234 binding to CEA in ELISA.
Binding of L12E2 to its corresponding C234 could be inhibited by CEA.
  
Potential use : L12E2 anti-idiotypic antibody could be used for:
1. Affinity purification of anti-CEA antibodies recognizing the same C234 epitope on CEA
2. Active immunization as a surrogate of CEA
3. Idiotype identification
  
Storage : Store at 4oC.
 
Stability : When reconstituted in aqueous solution IgG remains stable for six months if stored at 4oC. No considerable loss of activity was detected when stored at -20oC over a period of six months.
